二、實(shí)驗(yàn)室介紹

首都醫(yī)學(xué)科學(xué)創(chuàng)新中心姜漢杰課題組現(xiàn)面向國內(nèi)外招聘博士后與科研助理，課題組專注于利用化學(xué)生物學(xué)方法解碼蛋白質(zhì)翻譯后修飾在癌癥和其他疾病中的功能，并探索潛在的疾病治療策略。課題組堅(jiān)持科學(xué)點(diǎn)亮生活，鼓勵(lì)開放交流，支持成員學(xué)習(xí)成長，希望認(rèn)真研究同時(shí)認(rèn)真生活，立志為癌癥等疾病貢獻(xiàn)新的解題思路。如果你想一邊和志同道合的人們研究前沿科學(xué)，一邊享受實(shí)驗(yàn)室的快樂氛圍，趕緊看過來！

姜漢杰研究員畢業(yè)于北京大學(xué)，本碩階段的導(dǎo)師為蒲小平教授，于2022年在約翰霍普金斯大學(xué)獲得藥理學(xué)與分子科學(xué)的博士學(xué)位，師從PhilipCole教授，期間主要研究泛素連接酶的功能調(diào)節(jié)和底物識(shí)別機(jī)制，之后繼續(xù)在哈佛醫(yī)學(xué)院/布萊根婦女醫(yī)院完成了博士后訓(xùn)練，應(yīng)用化學(xué)生物學(xué)手段探究泛素連接酶的催化機(jī)制。姜漢杰研究員于2025年2月加入首都醫(yī)學(xué)科學(xué)創(chuàng)新中心。

三、主要研究方向

1.開發(fā)和應(yīng)用蛋白質(zhì)半合成、分子探針等化學(xué)方法，研究酶及細(xì)胞信號(hào)通路中的蛋白質(zhì)磷酸化、泛素化等翻譯后修飾。

2.探索蛋白質(zhì)泛素及類泛素修飾的調(diào)控機(jī)制以及其在疾病發(fā)生中的病理機(jī)制，鑒別新的治療靶點(diǎn)。

3.開發(fā)靶向泛素化的合成生物學(xué)工具，為基礎(chǔ)研究和疾病治療提供新方法。

四、主要成果

1.發(fā)現(xiàn)HECT家族E3連接酶末端羧基參與泛素轉(zhuǎn)移的催化機(jī)制。

2.揭示HECT家族E3連接酶中的自抑制和激活機(jī)制。

3.開發(fā)特異性的蛋白質(zhì)N端標(biāo)記方法。

Laboratory Introduction

The Jiang lab at CIMR now has open positions for postdoctoral fellows and research assistants. The lab is dedicated to using chemical biology approaches to decode the functions of protein post-translational modifications (PTMs) in cancer and other diseases, with a focus on advancing innovative therapeutic strategies. Dr. Jiang graduated from Peking University with Bachelor’s and Master’s degrees under the supervision of Dr. Xiaoping Pu. He received his Ph.D. degree in Pharmacology and Molecular Sciences from the Johns Hopkins University with Dr. Philip Cole in 2022, where he studied the regulation mechanisms and substrate selectivity of HECT family E3 ligases. He then completed his postdoc training in the Cole lab at Harvard Medical School and Brigham and Women’s Hospital, where he focused on illustrating the catalytic mechanisms of HECT E3 ligases using chemical biology approaches. Dr. Jiang joined CIMR faculty in February, 2025.

Major Research Projects

a. Developing and applying chemical methods, such as protein semisynthesis and molecular probes, to investigate protein ubiquitination, phosphorylation, and other PTMs in key enzyme and cellular signaling pathways.

b. Exploring the regulatory mechanisms of ubiquitin and ubiquitin-like modifications and their pathological roles in disease progression to identify novel therapeutic targets.

c. Designing and optimizing synthetic biology tools for targeted ubiquitination to provide innovative methods for basic research and translational medicine.

Major Contributions

a. Discovered the catalytic role of terminal carboxylate involvement in ubiquitin transfer by HECT E3 ligases.

b. Uncovered the autoinhibition and activation mechanisms in HECT E3 ligases.

c. Developed a specific protein N-terminal labeling method.

Postdoctoral Researcher

Qualifications

a. Have obtained or be about to obtain (within the past three years) a Ph.D. in life science, chemistry, pharmacology or a related field. Candidates with a strong background in fields, such as biochemistry and molecular biology, pharmacology, protein design or targeted-based drug development are preferred.

b. Highly responsible, honest with a positive mindset, and unafraid of trying new approaches.

c. Capable of conducting research independently.

Research Assistant

Qualifications

a. Bachelor’s degree or above in life sciences, chemistry, medicine, or a related field.
